ISOLASI DAN IDENTIFIKASI BAKTERI GRAM POSITIF KOKUS PADA KASUS EAR MITES KUCING DOMESTIK (Felis domesticus) DI KECAMATAN SYIAH KUALA KOTA BANDA ACEH

ABSTRAK
Penelitian ini bertujuan untuk mengisolasi dan mengidentifikasi bakteri Gram positif kokus yang diduga menjadi penyebab pada kasus ear mites kucing domestik di Kecamatan Syiah Kuala Kota Banda Aceh. Sampel yang digunakan berasal dari 10 ekor kucing domestik yang positif menderita ear mites. Hasil swab liang telinga kucing dimasukan ke media Nutrient Broth (NB) dan diinkubasikan pada suhu 37°C selama 24 jam, selanjutnya menggunakan osse steril dilakukan penanaman pada media Manitol Salt agar (MSA) dan media Plate Agar Darah (PAD) dengan teknik goresan T lalu diinkubasikan kembali selama 24-48 jam. Morfologi koloni yang tumbuh terpisah diamati bentuk, ukuran, warna dan elevasi sebelum dilakukan pewarnaan Gram. Uji biokimia (Manitol dan Glukosa) dan Uji katalase dilakukan pada bakteri yang berbentuk kokus. Hasil penelitian ini dari 10 sampel dinyatakan 8 sampel (80%) penyebab infeksi sekunder pada kasus ear mites adalah bakteri Gram positif dari kelompok Staphylococcus sp dan 2 sampel (20%) dari kelompok Streptococcus sp. Dari 8 sampel yang terinfeksi bakteri kelompok Staphylococcus, 3 sampel (37,5%) merupakan infeksi tunggal Staphylococcus aureus dan 2 sampel (20%) merupakan infeksi tunggal Staphylococcus epidermidis, selain itu terdapat infeksi ganda antara Staphylococcus aureus dan juga Staphylococcus epidermidis sebanyak 2 sampel (20%) dan infeksi ganda Staphylococcus aureus dengan Micrococcus sp sebanyak 1 sampel (12,5%).

ABSTRACT
The objectives of this study was to isolate and identify the Gram positive coccus bacteria which were suspected as one of the cause of secondary infection in ear mites case of domestic cats in Subdistric of Syiah Kuala, Banda Aceh. In this study, 10 of domestic cats suffer from ear mites were used. The swab ear canal from each cat was inoculated in Nutrient Broth (NB) and incubated at 37°C for 24 hours. Then, using steril osse bacteria were cultured in Manitol Salt Agar (MSA) and Blood Agar Plate (BAP) with T streak method and incubated for 24-48 hours. The morphology coloni of bacteria were observed for shape, size, colour and elevation before Gram staining procedure was applied. Biochemical (manitol and glucose) and catalase test then conducted for the colonies which were identified as coccus in shape. The result of this study showed, 8 samples (80%) out of 10 samples were positive infected by the group of Staphylococcus sp and 2 samples (20%) are positive infected by the group of Streptococcus sp. Frome those 8 samples infected by the group of Staphylococcus sp, 3 samples (37,5%) only infected by Staphylococcus aureus and 2 samples (25%) only infected by Staphylococcus epidermidis while the others 2 samples (25%) were infected by both Staphyloccocus aureus and Staphylococcus epidermidis and only 1 samples (12,5%) was infected by Staphylococcus aureus and Micrococcus sp. It can be concluded that the group of Gram positive bacteria was the one of the cause of secondary infection in the cats by ear mites.
